The cross-linking of proteins caused by aldehydes is reversible. Under certain conditions, such as high temperature or protease hydrolysis, the original conformation of the protein can be restored. This process is called antigen retrieval. Antigen retrieval is a key step that cannot be ignored in the process of immunohistochemistry. There are many antigen retrieval methods. Different methods can be selected according to the characteristics of tissue types. Some are suitable for thermal repair, and some are suitable for enzymatic repair. Before the heat-induced antigen retrieval method appeared, the epitope retrieval induced by trypsin, proteinase K, pepsin and other enzymes was the most commonly used method.

In the experimental steps of immunohistochemistry or immunofluorescence, in the antigen retrieval step, directly add the pepsin antigen retrieval solution to the tissue slides to cover the tissue, or soak the slides in the repair solution and incubate at 37°C for 10-30 mins. The specific time is adjusted according to the repair effect of the slides .
1. The pepsin antigen retrieval solution should be restored to room temperature before use.
2. In order to obtain the best experimental results, the optimal incubation time for antigen retrieval is recommended to be pre-experimented based on different samples and target proteins.
3. The pepsin antigen retrieval solution is acidic, which may cause poor tissue morphology on the slides. Please pay attention to the incubation time.
