Introduction:

This product is used to remove Mycoplasma cells. It is a combination reagent of quinolones, tetracyclines and macrolides. It can effectively remove Mycoplasma cells by inhibiting DNA replication and protein synthesis of Mycoplasma. Through optimization and adjustment, this product not only has low cytotoxicity and fast removal rate of mycoplasma, it can completely remove the mycoplasma contained in cultured cells in about 7 days. After replacing the normal medium, the cells are in good condition and there is no re-positive phenomenon in continuous multi-generation culture.

Storage and transportation

Transport with wet ice; store at -20°C to avoid repeated freezing and thawing. The validity period is 12 months.

Steps

 

1. Prepare medium A: Take 1 mL of SweMyco Elimination Reagent-1 (100×) and add it to 100 mL of cell culture medium (with serum, without double antibodies), and mix thoroughly;

 

2. Prepare medium B: Take 1 mL of SweMyco Elimination Reagent-2 (100×) and add it to 100 mL of cell culture medium (with serum, without double antibodies), and mix thoroughly;

 

3. When the mycoplasma-infected cells are subcultured or changed, first use medium A for routine cell culture for 4 days; then change medium B for routine cell culture for 3 days;

 

4. After one round of culture (7 days), inoculate a part of the cells into a 6-well plate (or other culture vessel) and replace it with a complete medium without SweMyco Elimination Reagent. The remaining cells continue to use medium A and B for cell culture alternately;

 

5. The cells in the 6-well plate are cultured in medium without SweMyco Elimination Reagent for 2-3 days, and when the confluence of the cells reaches 90% or more, the mycoplasma detection is performed (G1900, G1901, G1902 are recommended);

 

6. According to the test results, perform the following operations:

 

a) The result is positive: continue to repeat steps 3-5 until the test result is negative.

 

b) The result is negative: The cell group containing SweMyco Elimination Reagent medium is replaced with a normal complete medium, and mycoplasma detection is performed after a period of incubation. If the retest result is negative, the removal is complete.

 

Precautions

 

1. This product has been tested for removal in a variety of mycoplasma-infected cell lines, but different cell lines have different sensitivity to the reagent. If the cytotoxicity is too large or the removal effect is not good, the product concentration can be adjusted or extended appropriately. Clear time.

 

2. There are many ways to infect cells with Mycoplasma. It is recommended to add mycoplasma prevention reagents in daily culture (G4009 is recommended).

 

3. Cells that have been determined to be mycoplasma contaminated, try to use a separate ultra-clean table during operation to avoid cross-contamination.

 

4. For your safety and health, please wear lab coats and disposable gloves for operation.
