Product Description


The Recombinant SUMO Protease (Small ubiquitin-related Modifier Protease) is a commonly used UBL-specific Protease (UBL-specific Protease 1, Ubiquitin-like protein-specific protease 1 (ULP1) can recognize the tertiary structure of SUMO (Small ubiquitin-like Modifier) protein with high specificity. Hydrolyze the post-Gly-Gly peptide bond in the X-Gly-Gly-X (Except Pro) peptide at the carboxy-terminal of SUMO protein, so that the target protein released after cleavage of the fusion protein by this protease does not contain excess amino acids.The optimum pH and temperature for Sumo Protease digestion were 8.0 and 30℃.It has good stability and high enzyme activity in pH range 6.0-10.0, temperature range 2-30℃, ionic strength range 0-400 mM NaCl.In practice, in order to preserve the structure and biological activity of the target protein as much as possible, it is recommended to use Recombinant Sumo Protease digestion at 4℃overnight, and at the same time, adding appropriate concentration of DTT (0.5-2 mM) into the digestion system can significantly improve the digestion efficiency.

Application: It is commonly used to remove the SUMO tagged protein from fusion proteins.The protease itself carries the HIS label and binds to the nickel column.

Source: Saccharomyces cerevisiae cysteine protease, recombinant expression of Escherichia coli, with HIS label.

Enzyme activity definition: one unit of enzyme activity can be used to cut more than 85% of 2 μg of substrate (containing SUMO labeled protein) within 1 h at 30℃, pH8.0.

Inactivation or inhibition: Imidazole higher than 150 mM will affect the digestion efficiency of Recombinant SUMO Protease

Purity and concentration: SDS-PAGE test purity ≥95%, 10 U/μL.
