Collagenase is a proteolytic enzyme, commonly used to digest extracellular matrix proteins. As a kind of endopeptidase, collagenase specifically recognizes the Pro-X-Gly-Pro sequence (frequently occurs in collagen and rarely in other proteins) and cleaves amino acids (X) and glycine (Gly) The peptide bond between.


Commercial collagenase is derived from Clostridium histolyticum (Clostridium histolyticum), which is a crude enzyme extract. It is divided into type I collagenase, type II collagenase, type III collagenase, and type IV collagen due to the different components contained in it. Enzymes, type V collagenase, etc., have different uses according to their components and degradation characteristics.


Type I collagenase contains a relatively uniform amount of multiple enzyme activities (including collagenase, casein, clostridial protease, trypsin, etc.), and is usually used for the preparation of primary cells from epithelial tissue, liver, lung, fat, and adrenal tissue.


Type II collagenase contains higher clostridial protease activity, and is usually used for the preparation of primary cells from tissues such as heart, bone, liver, thymus, salivary gland and cartilage.


Type III collagenase has low secondary protease hydrolysis activity and is often used in the preparation of primary cells of breast tissue.


Type IV collagenase contains low pancreatin activity and is usually used to prepare islet cells and myeloid cells.


Type V collagen is a partially purified type with higher collagenase and caseinase activity and lower pancreatin level, which is suitable for preparing pancreatic islets.


This product is type I collagenase.

The following is for reference only: Use buffers containing calcium and magnesium ions such as HBSS, EBSS, etc. as a solvent to prepare 100× or 1000× storage solution, and then filter and sterilize with a low-protein adsorption filter (such as PVDF filter), and pack into Store small aliquots at -20°C to avoid repeated freezing and thawing. Thaw on ice before use, and then dilute with the corresponding buffer according to the required working concentration. The recommended working concentration range of collagenase is 50-200U/mL or 0.5-2.5 mg/mL.
