Van Gieson (VG) is used in collagen fibers staining. When using combined solutions of picric acid and acid fuchsin, the small molecules of picric acid penetrate all of the tissues rapidly, but are only firmly retained in the close textured, red blood cells and muscle. The larger molecules of acid fuchsin displaces picric acid molecules from collagen fibres, which have larger pores, and allow the larger molecules to enter. In this method, collagen fibers show red by acid fuchsin and muscle fibers show yellow by picric acid.
1.Deparaffinize and hydrate to water.
2.VG stain: Mix saturation picric acid solution and acid magenta solution in 9:1 to generate VG staining solution. Rinse slides in this solution for 3 min, wash in tap water quickly. Dehydrate quickly in 3 changes of absolute alcohol.
3.Clear in xylene for 1-5 min and mount with resin mounting medium.
4.Microscopy determination and collect images for analysis.
Collagenous fibers are red while other elements are yellow.
1.Stained slides easily fade out, please collect images as soon as possible as needed.
2.VG easily fade out when wash in water, please manage the times of wash and dehydrate.
